Pap stain is a universal stain used for gynecologic and non-gynecologic cytology smear. It is mainly used for oral and cervical cancer screening in asymptomatic population and in the follow up of patients with cancer. Pap test has decreased incidence of cervical cancer by 70% in developed countries.
The PAP stain is a polychromatic counterstaining method consisting of stains such as Orange G 6 (OG6) and modified eosin azure (EA). The strength of the Pap stain is such that it results in: Well-stained nuclear chromatin.
The principle of pap stain Solution 1 has Hematoxylin stain, a basic nuclear stain. Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
Because the Papanicolaou stain was developed to document keratinization (the orange G dye component binds avidly to keratin), it is supe- rior to Romanowsky-type stains for this purpose. Identification of cytoplasmic keratinization may be essential for diagnosing squamous cell carcinomas.
The first staining solution contains haematoxylin which stains cell nuclei. Papanicolaou used Harriss hematoxylin in all three formulations of the stain he published. The second staining solution (designated OG-6), contains Orange G in 95% ethyl alcohol with a small amount of phosphotungstic acid.
EA50 is a counter stain designed to stain the cytoplasm of cells derived from the human body. The stained cell sample is used to facilitate the diagnosis of the pathological state, and is intended for in-vitro examination of cellular deformity. PRINCIPLE OF METHOD: Papanicolaou staining (Pap.
Basic Procedure for Papanicolaou Staining (Pap stain) Fix the smear with 95% Ethanol 15 minutes. Rinse in tap water. Add the Harris Hematoxylin dye for 1-3 minutes. Rinse in tap water or Scotts tap water. Dip the preparation in 95% Ethanol 10 dips. Add orange G-6 stain for 1.5 minutes. Dip in 95% Ethanol 10 dips.
Procedure of PAP stain Fix the smear immediately in 95% alcohol for 5-15 min. Take the smear to water (Hydration): 6 dips in each for the following solution of ethanol 80% (v/v) , 70%(v/v)and 50%(v/v) respectively. Rinse gently in distilled water. Stain in diluted Harriss hematoxylin for 2 to 3 minutes.
The conventional Pap stain has 24 solutions, of which three are acid dyes, six water, one is chemically defined bluing agent, eight 95% ethanol, three absolute alcohol and three xylene Gill, 2006.
The classic form of Pap stain involves five dyes in three solutions: A nuclear stain, haematoxylin, is used to stain cell nuclei. First OG-6 counterstain. The Orange G is used to stain keratin.
