When these cells are active, there is an increase in blood flow and blood oxygen in the surrounding area. BOLD imaging uses magnetic fields to measure this change in oxygenation. This signal is often used to infer the activity of brain cells and, thus, which brain areas are involved in a particular task.
The BOLD signal is primarily determined by the change in paramagnetic deoxygenated hemoglobin, which results from combination of changes in oxygen metabolism, and cerebral blood flow and volume.
fMRI BASICS. The basic principle underlying fMRI is that it studies physiological processes rather than structural processes. It is based on the BOLD, blood oxygenation level dependent, effect. BOLD exploits the fact that deoxyhemoglobin creates magnetic field gradients which decrease MR signal.
BOLD contrast is generated by changes in local deoxyhemoglobin, and depending on the field strength and scanning sequence used, the relative contributions of smaller venules and capillaries can be enhanced. For most field strengths used in humans, the signal primarily arises in the veins and capillaries.
Blood Oxygenation Level Dependent (BOLD) imaging is a technique that is commonly used for measuring brain activity in humans using magnetic resonance imaging (MRI).
The blood-oxygen-level-dependent (BOLD) signal, detected in fMRI, reflects changes in deoxyhemoglobin driven by localized changes in brain blood flow and blood oxygenation, which are coupled to underlying neuronal activity by a process termed neurovascular coupling.
The BOLD Response: A Fundamental Issue When neurons fire or increase their firing rate, they draw on oxygen and various nutrients. The circulatory system of the brain reacts by sending the region that just fired more highly-oxygenated blood than is needed.
Characteristic features of the BOLD response are a delay of a few seconds after the start of the stimulus, a ramp of approximately 6 s up to a plateau, and a post-stimulus undershoot before the signal returns to baseline. The average response in Fig. 16.1 is fairly typical of BOLD responses to a number of stimuli.
