DNA barcoding has three main steps: DNA extraction, PCR amplification, and DNA sequencing and analysis (Figure 1).
BOLD was originally developed by Ratnasingham, a computer scientist and CBG Director, Prof. Paul Hebert, to support the generation and application of data from DNA barcodes a method that uses short, standardized genetic markers to identify species.
DNA barcoding involves sequencing a short fragment of the mitochondrial cytochrome c oxidase subunit I (COI) gene, DNA barcodes, from taxonomically unknown specimens and performing comparisons with a library of DNA barcodes of known taxonomy.
A good DNA barcode should have low intra-specific and high inter-specific variability and possess conserved flanking sites for developing universal PCR primers for wide taxonomic application.
The Barcode of Life Data System (commonly known as BOLD or BOLDSystems) is a web platform specifically devoted to DNA barcoding. It is a cloud-based data storage and analysis platform developed at the Centre for Biodiversity Genomics in Canada.
GenBank (1) is a comprehensive public database of nucleotide and protein sequences with supporting bibliographic and biological annotation, built and distributed by the National Center for Biotechnology Information (NCBI), a division of the National Library of Medicine (NLM), located on the campus of the US National
BOLD was originally developed by Ratnasingham, a computer scientist and CBG Director, Prof. Paul Hebert, to support the generation and application of data from DNA barcodes a method that uses short, standardized genetic markers to identify species.
BOLD is a curation tool that also stores sequences, while GenBank is just a sequence repository. Many sequences are duplicated between databases, as all BOLD sequences are automatically submitted to GenBank (denoted by the key term BARCODE) and BOLD periodically mines barcode sequences from GenBank [7,8].
PCR Cloning Combined With DNA Barcoding Enables Partial Identification of Fish Species in a Mixed-Species Product. DNA barcoding is a valuable tool for regulatory identification of fish species; however, it does not perform well when multiple species are present within the same food product.
The Barcode of Life Data System (bold) is an informatics workbench aiding the acquisition, storage, analysis and publication of DNA barcode records. By assembling molecular, morphological and distributional data, it bridges a traditional bioinformatics chasm.
