if Iamp;#39;m say Iamp;#39;m shooting handheld and Iamp;#39;ve been in these positions where Iamp;#39;ve done this beautiful handheld operation and then everyone seems fine with it on the day and then when we get to the final film the emotion that where weamp;#39;re going with all this has now changed a little bit in the edit whether itamp;#39;s pacing whether the character Arc is a little different and the the director comes to me and says you know what that that handheld is just a little too intense itamp;#39;s a little too shaky so Iamp;#39;m like okay well I would have loved to have known that when I was shooting it so um but if itamp;#39;s too shaky then what weamp;#39;re gonna have to do is weamp;#39;re gonna have to zoom in on the image so we can give it the ability to stabilize so now what Iamp;#39;ve done across the board anytime Iamp;#39;m shooting handheld I shoot 5K but frame for 4K what that does is it gives me 25 around the edges to be able to stabilize okay

What Does Quantitation Mean? Quantitating a western blot refers to the measurement of the signal emitted by your protein band(s) of interest. The signal intensity of the band is directly proportional to the concentration of your target protein.

ASC specks are micrometer-sized structures formed by the inflammasome adaptor protein ASC (apoptosis-associated speck-like protein containing a CARD). ASC specks are characteristic of inflammasome activation. The inflammasome adapter ASC is localized mainly in the nucleus of macrophages and monocytes.
This type of saturation can affect any Western blot assay, regardless of the detection chemistry or imaging method [11,15,77]. Signal saturation occurs when the intensity of a strong band exceeds the capacity of the detection chemistry or imaging system.
Dynamic and Linear Range in Western Blot Imaging Saturation is the point at which the sensor pixels can no longer convert additional incoming photons to electrons. Imaging software will identify these pixels and usually display them in a different color.
An important problem that usually arises in signal processing is the signal saturation (Swanson, 2000). An excessive intensity of the analog signal from a sensor may yield an analog signal which is cut off (or saturated) at the range limits of the analog-to-digital converter, as shown in Figure 4.
Western blots assess how much of a particular protein is present in a sample. The thicker or darker the band, the more of the protein is present in the sample.
A western blot is a laboratory method used to detect specific protein molecules from among a mixture of proteins. This mixture can include all of the proteins associated with a particular tissue or cell type.
