These filter papers are smooth and suitable for use with alcohol and other organic solvents involved in protein transfer and nucleic acid blotting. They also allow for a uniform flow of buffer through the gel to the transfer membrane in a blotting sandwich.
The standard transfer buffer for western blots, called Towbin buffer, is 25 mM Tris, 192 mM glycine, pH 8.3 usually with 20% methanol (vol/vol). Sometimes SDS is added to this buffer, generally in the range of 0.1 to 0.25%.
What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
Tris Glycine Transfer Buffer with 20% methanol. 1X Transfer Buffer should be pH 8.3 before addition of SDS or methanol.
Methanol is only necessary when nitrocellulose is used. If using PVDF, methanol can be removed from the transfer buffer altogether, and you just need to activate the PVDF with methanol before assembling the gel/membrane sandwich ing to the right order.
SDS in the gel and in the SDS-protein complexes promotes elution of the protein from the gel but inhibits binding of the protein to membranes. In cases where certain proteins are difficult to elute from the gel, SDS may be added to the transfer buffer to improve transfer.
Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk diluted in TBST to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available.
