SDS-PAGE. In SDS-PAGE, the gel is cast in a buffer containing sodium dodecyl sulfate (SDS), an anionic detergent. SDS denatures proteins by wrapping around the polypeptide backbone.
SDS, DTT, and heat are responsible for the actual denaturation of the sample.
The anionic detergent, sodium dodecyl sulphate (SDS), in combination with heat and sometimes a reducing agent is used to denature proteins prior to electrophoretic separation in a process known as SDS PAGE.
SDS is a detergent, an anionic (negatively charged) surfactant (compound that lowers surface tension). In the case of proteins, SDS disrupts the non-covalent bonds in protein molecules.
Factors affecting protein migration include: Gel composition and strength (weight percentage of polymer) Electric field strength. Temperature. Protein size, shape, and charge which are in turn affected by. Buffer pH, ion type, and concentration. Addition of detergents or reducing agents.
Typically, you will boil your protein samples in the loading buffer (containing Tris-HCl, SDS, bromophenol blue, glycerol, and ?-me) before loading them in your gel. This helps to completely denature the proteins and also helps with physically loading the gel.
Sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) is commonly used to obtain high resolution separation of complex mixtures of proteins. The method initially denatures the proteins that will undergo electrophoresis.
SDS-PAGE separates proteins primarily by mass because the ionic detergent SDS denatures and binds to proteins to make them uniformly negatively charged. Thus, when a current is applied, all SDS-bound proteins in a sample will migrate through the gel toward the positively charged electrode.
