[Music] in this video we will show you how to prepare a western blot membrane for chemiluminescent detection after antibody incubation and washing the membrane is ready to be incubated in enhanced chemiluminescent or ecl substrate prepare enough ecl substrate for the blot for a mini-sized membrane use about seven milliliters of substrate increase the volume to 12 milliliters if using a medium-sized membrane these volumes ensure uniform coverage and optimal results when too little substrate is used residual wash buffer is more likely to adversely affect the performance of the horseradish peroxidase enzyme although any small container will work for substrate incubation a clean sheet protector is recommended as it reduces the volume of substrate needed sheet protectors are also inexpensive and disposable incubate for five minutes to ensure the substrate has enough time to uniformly coat the membrane and displace the wash buffer membranes can also be kept inside a sheet protector for imag

The Ponceau stain will identify bubbles which have interfered with protein transfer, and appear as blank circles among the transferred proteins. Make sure to expel air from between the gel and membrane before transfer.
The answer is yes: western blotting Coomassie-stained proteins can be done, but it's not a simple or efficient process. As you know, there are two types of Coomassie stains – “classical” and “colloidal”.
When proteins bind to Coomassie blue in acid solution their positive charges suppress the protonation and a blue colour results. The binding of the dye to a protein causes a shift in the absorption maximum of the dye from 465 to 595 nm and it is the increase in absorbance at 595 nm that is monitored.
Coomassie brilliant blue R250 and amido black 10B are more sensitive than Ponceau S2. However with Coomassie brilliant blue R250 the background staining is high and also this stain is not removed easily.
The answer is yes: western blotting Coomassie-stained proteins can be done, but it's not a simple or efficient process. As you know, there are two types of Coomassie stains – “classical” and “colloidal”. Proteins stained by one of these two methods will behave differently if you try to blot them afterwards.
Ponceau S membrane staining is a nondestructive, reversible method to stain and detect proteins on nitrocellulose and PVDF membranes. The stain has minimal nonspecific interactions with the membrane surface and provides a reliable method for visualization of protein transfer to a membrane.
Note that complete destaining is only achieved in the subsequent blocking step and that the Ponceau S stain will not interfere with either blocking or antibody binding steps.
Oriole Fluorescent Gel Stain is an easy-to-use, fast, sensitive, and highly linear protein gel stain for visualization and quantitation of proteins in SDS-PAGE gels. The simple protocol requires a one-step staining process without prior fixing.
Amido black is used to stain proteins on blot transfer membranes. Transferred proteins (>50 ng/band) appear as dark blue bands on a light blue background. Amido black has a sensitivity similar to that of Coomassie blue, but it stains faster.
there can be two reasons. One: there is not enough amount to be detected by coomassie staining on SDS PAGE. but because western detects very low amount of proteins you can see it on western blot. the amount of it present is simply less than the detection limit of coomassie.
