Hi Im Megan today were going to stain a blood smear so that we can look at it under the microscope. Now here in this hospital we use the right stain and so the times that im going to give you for leaving the slide in each jar are consistent for what we use at this hospital - it may be a little bit different where you are. Its also important to make sure that your stain jars stay clean and that they stay free of debris. What we do here is we have a set of stain for our CBCs and cytologies, we have one for urine and we have one for fecals. And thats so that we dont cross contaminate and start to get really interesting results on our different cytologies and UAs so we make sure were not mixing things. We also empty the stain out of these jars, clean the jars and let them dry at least once a week so thats going to vary also depending on where you are and how often you use them but you can get things that grow in your stain that will show up on your blood films and its going to y

How to stain PVDF membrane? Soak blott after D.I. water rinse with methanol. Stain PVDF membrane with 0.2% Amido Black in 40% MeOH for 30 seconds to one minute. Destain in D.I. water with multiple changes until bands are clear and low background. Cut out band of interest.
Coomassie brilliant blue stain is easily available and is the most commonly used protein stain.
Silver staining is the most sensitive colorimetric method for detecting total protein. The technique involves the deposition of metallic silver onto the surface of a gel at the locations of protein bands. Silver ions (from silver nitrate in the staining reagent) interact and bind with certain protein functional groups.
Stain-Free imaging technology utilizes a polyacrylamide gel containing a proprietary trihalo compound to make proteins fluorescent directly in the gel with a short photoactivation, allowing the immediate visualization of proteins at any point during electrophoresis and western blotting.

INTRODUCTION. Coomassie Blue R250 permanently stains membrane-bound proteins and is compatible with PVDF and nitrocellulose membranes, but it is incompatible with nylon membranes. This technique is relatively insensitive, with a detection limit of ~1.5 μg of protein.
Materials Place blot transfer membrane in a plastic box. ... Rinse with water four times for 5 min each. Stain with SYPRO Ruby protein blot stain for 15 min. ... Rinse with water four to six times for nitrocellulose and two to three times for PVDF for 1 min each, then air-dry.
What Is Stain-Free Technology? Mini-PROTEAN TGX Stain-Free Precast Gels are based on the long-shelf-life TGX (Tris-Glycine eXtended) formulation and include unique trihalo compounds that allow rapid fluorescent detection of proteins with Bio-Rad stain-free imaging systems.
The answer is yes: western blotting Coomassie-stained proteins can be done, but it's not a simple or efficient process. As you know, there are two types of Coomassie stains – “classical” and “colloidal”. Proteins stained by one of these two methods will behave differently if you try to blot them afterwards.
Amido black is used to stain proteins on blot transfer membranes. Transferred proteins (>50 ng/band) appear as dark blue bands on a light blue background.
