Coomassie Blue staining is approximately 50-fold less sensitive than silver staining, however due to its simplicity binding Coomassie Blue is preferred.
Ponceau S membrane staining is a nondestructive, reversible method to stain and detect proteins on nitrocellulose and PVDF membranes. The stain has minimal nonspecific interactions with the membrane surface and provides a reliable method for visualization of protein transfer to a membrane.
Amido black is used to stain proteins on blot transfer membranes. Transferred proteins (>50 ng/band) appear as dark blue bands on a light blue background. Amido black has a sensitivity similar to that of Coomassie blue, but it stains faster.
The dye rapidly stains proteins on membranes pink or light red for easy visual inspection of protein transfer. The staining is reversible and compatible with subsequent Western blotting [21]. Incubate membrane in Ponceau S solution for 5-10 minutes at room temperature (RT).
A Ponceau S stain is useful because it does not appear to have a deleterious effect on the sequencing of blotted polypeptides and is therefore one method of choice for locating polypeptides on western blots for blot-sequencing.
Fluorescent staining is a popular alternative to traditional staining procedures, mainly because it is more sensitive than Coomassie staining, and often as sensitive as silver staining.
The Ponceau stain will identify bubbles which have interfered with protein transfer, and appear as blank circles among the transferred proteins. Make sure to expel air from between the gel and membrane before transfer.
Coomassie brilliant blue R250 and amido black 10B are more sensitive than Ponceau S2. However with Coomassie brilliant blue R250 the background staining is high and also this stain is not removed easily.
Western blot allows the transfer of proteins separated on a gel (SDS-PAGE) to a sticky membrane. Protein transfer is facilitated by electric current, negatively charged proteins and supporting reagents such as filter paper, transfer buffer, and a nitrocellulose or PVDF membrane.
Ponceau S detects protein levels at 200 ng and higher, is compatible with PVDF, nitrocellulose, or nylon membranes. The traditional Coomassie Brilliant Blue is only compatible with PVDF membranes, but can detect protein levels at 50 ng and higher.
