Western blot allows the transfer of proteins separated on a gel (SDS-PAGE) to a sticky membrane. Protein transfer is facilitated by electric current, negatively charged proteins and supporting reagents such as filter paper, transfer buffer, and a nitrocellulose or PVDF membrane.
The optimal fixation method for PVDF membranes, was acetone treatment at room temperature followed by sample heating at 100 C, both for 30 min. In the case of nitrocellulose membranes, the optimal fixation method was a combination of incubation in 50% methanol solution and heating at 100 C for 30 min each.
Western blotting is a laboratory technique used to detect a specific protein in a blood or tissue sample. The method involves using gel electrophoresis to separate the samples proteins. The separated proteins are transferred out of the gel to the surface of a membrane.
Tetrazolium is an indicator dye that is colorless in its oxidized form, but pink when it is reduced. When microorganisms metabolize carbon compounds they make waste products that serve as reducing agents, also called reductants or electron donors, that will reduce tetrazolium, turning it pink.
The size of the target protein should be considered when choosing transfer conditions. Smaller proteins will transfer out of the gel faster and may actually transfer through the PVDF membrane into the filter papers beyond. PVDF with a smaller pore size can be used for small proteins and peptides.
Western blotting is frequently used for the confirmatory medical diagnosis of infectious diseases such as Lyme disease, HIV infection, bovine spongiform encephalopathy (BSE), hepatitis C infection, syphilis, inflammatory muscle conditions such as myositis, and certain autoimmune disorders (e.g., paraneoplastic disease)
Tetrazolium staining is a popular method for early detection of myocardial infarction in a whole heart. This technique relies on the ability of dehydrogenase enzymes and cofactors in tissue to react with tetrazolium salts to form a pigment.
2,3,5-Triphenyltetrazolium chloride (TTC) staining is a commonly used method to determine the volume of the cerebral infarction in experimental stroke models.
This routine type of Western blot is used to either test for endogenous cellular expression of a target protein, or to examine transfected cell lines to see if expression has been conferred by the introduction of a DNA construct .
Silver staining is the most sensitive colorimetric method for detecting total protein. The technique involves the deposition of metallic silver onto the surface of a gel at the locations of protein bands. Silver ions (from silver nitrate in the staining reagent) interact and bind with certain protein functional groups.
