[Music] stain-free technology is a unique tool that enables researchers to speed up their western blotting experiments and achieve better results by eliminating staining steps and providing checkpoints during electrophoresis and transfer users can ensure that conditions are optimized prior to blotting in addition the unique properties of the stain-free compound enable researchers to perform total protein normalization which is far more accurate than traditional housekeeping protein normalization particularly at higher protein concentrations because the stain-free compound is already embedded in the acrylamide gel you can visualize the resolved proteins with a stain-free enabled imager minutes after electrophoresis no protein stains such as kumasi blue or silver staining are needed saving you valuable time following a brief uv light activation stain-free compounds already present in the gel covalently bind protein molecules allowing them to be imaged repeatedly on the gel or on a membr

Coomassie brilliant blue stain is easily available and is the most commonly used protein stain.
The stains include anionic dyes (Coomassie brilliant blue), metal cations (imidazole-zinc), silver stain, fluorescent dyes, and radioactive probes.
The answer is yes: western blotting Coomassie-stained proteins can be done, but its not a simple or efficient process. As you know, there are two types of Coomassie stains classical and colloidal.
Visualization of protein bands is carried out by incubating the gel with a staining solution. The two most commonly used methods are Coomassie and silver staining. Silver staining is a more sensitive staining method than Coomassie staining, and is able to detect 25 ng protein per band on a gel.
Coomassie blue staining: Coomassie blue staining is the widely used method for staining SDS-PAGE gels.
Staining of proteins separated on gels provides the basis for determination of the critical properties of these biopolymers, such as their molecular weight and/or charge. Detection of proteins on gels and blots require stains.
The main disadvantage of Western blotting is that this technique requires a specific antibody to a target protein; thus many protein targets cannot be investigated because of the lack of specific antibodies.
Western blot allows the transfer of proteins separated on a gel (SDS-PAGE) to a sticky membrane. Protein transfer is facilitated by electric current, negatively charged proteins and supporting reagents such as filter paper, transfer buffer, and a nitrocellulose or PVDF membrane.
The most common method is staining with Coomassie blue, which after washing gives blue bands on a clear background. This technique can also be applied to isoelectric focusing gels.
Western blots can also be used to evaluate the size of a protein of interest, and to measure the amount of protein expression. This procedure was named for its similarity to the previously invented method known as the Southern blot.
