SDS-PAGE (sodium dodecyl sulfatepolyacrylamide gel electrophoresis) is a discontinuous electrophoretic system developed by Ulrich K. Laemmli which is commonly used as a method to separate proteins with molecular masses between 5 and 250 kDa.
Ideally, it is best to load 2 g per well of a purified protein or 20 g of a complex mixture like whole cell lysates if you are doing Coomassie stain only. Protein loading can be adjusted ingly for more sensitive stains like silver and fluorescent staining or when doing WB where you can do lower amounts.
Western blots can also be used to evaluate the size of a protein of interest, and to measure the amount of protein expression. This procedure was named for its similarity to the previously invented method known as the Southern blot.
Western blot is often used in research to separate and identify proteins. In this technique a mixture of proteins is separated based on molecular weight, and thus by type, through gel electrophoresis. These results are then transferred to a membrane producing a band for each protein.
To do this, researchers use a technique called densitometry to measure the relative amount of a specific protein for a given experimental sample on the blot by comparing it with a control or another protein.
By analyzing the intensity of the signal, you may determine whether the expression of the target protein in one sample has increased or decreased relative to another sample or control. Note: western blots are semi-quantitative so they are ideal for showing relative protein expression rather than an absolute quantity.
Chemiluminescent substrates emit light, which may be visualized using film or a CCD camera. Fluorescent western blotting uses a fluorescent dye molecule to provide a signal which is captured using an imager and may be used to facilitate the detection of multiple target proteins simultaneously.
