Sodium thioglycollate is a reducing agent which maintains a low oxygen tension in anaerobic media. A small amount of agar is added to impede diffusion of oxygen. Casein peptone and cystine supply nitrogenous and carbon compounds, while the sodium chloride maintains osmotic equilibrium.
Fluid Thioglycollate Medium (FTM) is primarily intended for the detection of anaerobic bacteria. However, it also enables the detection of aerobic bacteria.
The main application for FTM medium is in sterility testing of products within pharma manufacturing. The harmonised pharmacopoeia medium is typically used alongside Tryptic Soy Broth (TSB). As with most liquid medium, growth of microorganisms shows as turbidity in the otherwise clear liquid.
Fluid Thioglycollate Medium is a universal complex medium for the isolation and cultivation of fastidious anaerobic as well as for aerobic microorganisms. Fluid Thioglycollate Medium is used for sterility control of pharmaceutical products.
The mediums complex formulation includes a small amount of agar and resazurin. The agar controls the ingress of oxygen within the medium to maintain anaerobiosis in the lower part of the medium. The resazurin is an indicator dye which in the presence of oxygen turns pink.
What is the purpose of Thioglycollate media? To determine the oxygen requirement of a bacteria.
Thioglycolate broth is a multipurpose, enrichment, differential medium used primarily to determine the oxygen requirements of microorganisms. Sodium thioglycolate in the medium consumes oxygen and permits the growth of obligate anaerobes.
Aerobic and anaerobic bacteria can be identified by growing them in test tubes of thioglycolate broth: 1: Obligate aerobes need oxygen because they cannot ferment or respire anaerobically. They gather at the top of the tube where the oxygen concentration is highest.
