hi folks welcome back to learn from Dad Iamp;#39;m James today weamp;#39;re out in the garage and weamp;#39;re going to talk about hardware and specifically nuts and bolts and how do you figure out what size they are and now why do I care if I have a lawnmower or something in my house or on my vehicle and suddenly thereamp;#39;s a bolt missing or a nut missing you have to figure out what size it is that you can get the proper replacement the last thing you want to do is take a metric nut and an SAE or standard Bolt try to thread those together and run yourself into a problem so letamp;#39;s take a look at how do we size bolts and make sure that I can get the proper one if I have to do a replacement okay so letamp;#39;s take a look at some bolts and try to identify what the size of these are based on the three different parameters that you use to figure out a bolt now before you start measuring just like I showed in a previous video itamp;#39;s always good to figure out is this a

Nitrocellulose Membranes Pore sizes can be either 0.2 m or 0.45 m. The 0.45 m pore membrane is used for most targets because the 0.2 membrane is more efficient in binding smaller proteins. However, you can often switch when probing for ones that are less than 15 kDa.
By analyzing the intensity of the signal, you may determine whether the expression of the target protein in one sample has increased or decreased relative to another sample or control. Note: western blots are semi-quantitative so they are ideal for showing relative protein expression rather than an absolute quantity.
Chemiluminescent substrates emit light, which may be visualized using film or a CCD camera. Fluorescent western blotting uses a fluorescent dye molecule to provide a signal which is captured using an imager and may be used to facilitate the detection of multiple target proteins simultaneously.
Western blots can also be used to evaluate the size of a protein of interest, and to measure the amount of protein expression. This procedure was named for its similarity to the previously invented method known as the Southern blot.
Western blot is often used in research to separate and identify proteins. In this technique a mixture of proteins is separated based on molecular weight, and thus by type, through gel electrophoresis. These results are then transferred to a membrane producing a band for each protein.
To do this, researchers use a technique called densitometry to measure the relative amount of a specific protein for a given experimental sample on the blot by comparing it with a control or another protein.
Immobilon PVDF membranes offer excellent protein binding and high signal. PVDF Membranes Pore Size 0.2 M Detection methods Chemiluminescence, Colorimetric Binding capacity Goat IgG 448 g/cm2 BSA 340 g/cm26 more rows
To obtain linear signals with the majority of western blots, we recommend loading smaller amounts of protein sample between 1 and 10 g per well. To avoid under- or overloading samples, determine the protein concentration of each sample prior to electrophoresis with a compatible protein assay.
