Store at room temperature (20 to 25C) in original container protected from direct sunlight in a dry, cool and well- ventilated area. When properly stored, the reagent is stable for one year.
Edge Effect: When multiple lanes are run on a blot, the outer lanes may experience the edge effect. The edge effect occurs when proteins drift towards the center; luckily, this can be removed through the use of a loading control, fine-tuning the entire blotting process by correcting the variation in binding.
You can store it in TBST or blocking buffer at 4 degree. I would suggest incubation with primary antibody again before adding secondary antibody. Try not to do that for future works and its always best to proceed to secondary antibodies incubation after it.
Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk diluted in TBST to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available.
Put two pre-soaked blotting pads in the bottom of the electrode, then the first gel sandwich, with the MEMBRANE SIDE UP.
Generally, maximum blocking time should not exceed 2 hours at room temperature or proteins can be exchanged from the membrane.
What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
I would not recommend to keep the blot with primary antibody for more than 24hours. Treating the primary antibody for more than 24hrs would cause lot of unspecific bands and background.
