I wanted to ask you about tapping into the shadow side so sharing your darker size in your books how do you recommend the authors kind of tap those deeper places itamp;#39;s really really difficult like I wrote a blog post a couple of weeks ago it was called two paragraphs that was the title and what it was it was about the knowledge and and in that book there are two paragraphs following each other that are absolutely true from my heart the the real stuff that really took place in my real life that Iamp;#39;ve been trying to write about and I couldnamp;#39;t for 35 years and the whole rest of the book is just there to carry those two paragraphs so youamp;#39;ve got to be incredibly careful in using that real stuff because it it goes over into self-pity you totally lose objectivity on it and it needs to be it needs a stroke of genius I think in the sense of the concept of the story like whatamp;#39;s an example I donamp;#39;t know I canamp;#39;t think of an example off the top o

Patchy and uneven spots on the blot are usually caused by improper transfer. If there are air bubbles trapped between the gel and the membrane, it will appear darker on the film. It is also important to use a shaker for all incubation, so that there is no uneven agitation during the incubation. The appearance of white bands (ghost bands) on blots is usually caused by an intense localized signal, that completely exhausts the ECL reaction with a quick burst of light. Therefore, there is no light produced during development, and a white band occurs. This can also happen with prolonged incubation. What is a reason for so called ? ghost bands? to appear on the blots? Agrisera technical-blog 2023/03/30 Agrisera technical-blog 2023/03/30
While working with proteins in the lab, they should be kept on ice. Since proteins are generally more stable at colder temperatures, maintenance at low temperatures even for short duration is recommended. Typically, proteins are stored freeze-dried (lyophilized), frozen in appropriate buffers, or refrigerated at 4C.
For best results, use non-ionic or zwitterionic detergents (e.g., Tween 20, CHAPS). Adding a ligand to the protein solution before concentration will favor the population of proteins in their native state and reduce the exposure of hydrophobic patches in ligand-binding sites to prevent aggregation.
For reducing Western blotting, a reducing agent such as beta-mercaptoethanol or dithiothreitol (DTT) is often added to the sample buffer to break the disulfide bonds between cysteine residues and fully denature the proteins.
Loading too much protein leads to signal saturation in western blots, yet too little produces weak signals. This protocol describes an assay development experiment to determine the appropriate protein load for target and control detection prior to performing the actual western blot experiment. Sample degradation due to overheating or protease activity results in target breakdown and low target recognition by the antibody. For example, do not boil SDS-PAGE samples in SDS sample buffer, but rather heat them at 70C for 10 minutes to avoid proteolysis.
Here are some tips on how to handle and preserve your protein samples for Western blotting: Work quickly. Working quickly can minimize potential damage to your samples by simply allowing less time for them to become degraded. Keep everything cool. Use protease inhibitors.
