hi all hope youamp;#39;re doing well I wanted to shoot a quick first impression of this the Enzo bolt itamp;#39;s a hotly anticipated fountain pen that was part of a Kickstarter campaign itamp;#39;s a fully titanium fountain pen that uses a bolt action to extend and retract a retractable nib and Iamp;#39;ve used this now for about a week and I wanted to go ahead and give you my first impressions because many people are asking about it I did purchase it with the optional clip and Iamp;#39;ll show the installation process for this clip in the full review but overall I do like this clip quite a bit it has a nice flare out so itamp;#39;s easy to get in and out of fabrics and it is quite springy and functional in terms of the bolt action itself I found that the uh overall length to extend and retract the nib is a bit on the long side if you have medium or smaller hands you might find itamp;#39;s kind of a stretch but as you use it more and more it does become more second nature anoth

To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
Procedure Prepare lysis buffer by adding protease and phosphatase inhibitors. Dissect the tissue of interest on ice and weigh samples. Add the appropriate amount of ice-cold lysis buffer to the tissue sample and homogenize on ice. Centrifuge the sample at 10,000 g for 5 minutes to pellet cell/tissue debris.
This blot, as it is called, has an imprint of the bands of nucleic acid or protein that were in the gel (see figure at left). The transfer can be accomplished by diffusion or by using an electrical current to move the molecules from the gel onto the membrane.
For a routine Western blot, it is recommended to run the gel in reducing/denaturing conditions. For this, the lysate must be boiled in sample buffer at +95-100C (5 minutes) or at +70C (10 minutes). Then, samples can be immediately loaded on a gel or stored at -20C for later analysis.

The minimum recommended concentration is 0.1 mg/mL, optimal concentration is 15 mg/mL). Centrifuge for 510 min at 14,00017,000 g at 4C in a microcentrifuge. Gently remove the tubes from the centrifuge and place on ice, aspirate the supernatant and place in a fresh tube kept on ice. Discard the pellet.
