in this video weamp;#39;re going to talk about the theory and practice of western blotting so weamp;#39;re going to go through a little bit of why we do all the different steps that we do and then also talk about practically how to do them in the lab so the first thing i want to say about western blots is that they can be a little bit tricky just because they are so long they usually take two days to do with sort of the standard protocols and there are many many different steps and therefore many different points where something could go wrong and unfortunately with the western blot you donamp;#39;t really visualize your results until the very end and so if something does go wrong itamp;#39;s hard to know and that is why a lot of people have trouble when they first start doing these but if you go step by step and you sort of troubleshoot every step as you go it is a pretty straightforward protocol and once you have some practice i think itamp;#39;s very doable so just be very syst

For a routine Western blot, it is recommended to run the gel in reducing/denaturing conditions. For this, the lysate must be boiled in sample buffer at +95-100C (5 minutes) or at +70C (10 minutes). Then, samples can be immediately loaded on a gel or stored at -20C for later analysis.
Procedure Prepare lysis buffer by adding protease and phosphatase inhibitors. Dissect the tissue of interest on ice and weigh samples. Add the appropriate amount of ice-cold lysis buffer to the tissue sample and homogenize on ice. Centrifuge the sample at 10,000 g for 5 minutes to pellet cell/tissue debris.
Reduce and denature the samples by boiling the lysates in Laemmli Buffer at 95-100˚C for 5 minutes. Note: This step should only be skipped if the antibody datasheet recommends non-reducing or non-denaturing conditions.
Sample Preparation To a volume of protein sample (cell or tissue lysate), add equal volume of loading buffer. Boil the above mixture at 95 C for 5 min. Centrifuge at 16000 xg for 5 min. These samples can be stored at -20 C or may be used to proceed with gel electrophoresis.
Western Blot Western blotting is a laboratory technique used to detect a specific protein in a blood or tissue sample. The method involves using gel electrophoresis to separate the samples proteins. The separated proteins are transferred out of the gel to the surface of a membrane.
Western blot protocol Stage 1 - Sample preparation. Stage 2 - Loading and running the gel. Stage 3 - Transferring from the gel to the membrane. Stage 4 - Checking the success of transfer (optional) Stage 5 - Blocking and antibody incubation. Stage 6 - Detection. Stage 7 - Membrane stripping (optional) Stage 8 - Data analysis.
The Western blot test separates the blood proteins and detects the specific proteins (called HIV antibodies) that indicate an HIV infection.
Less sensitive than ELISA: Western blotting is less sensitive than ELISA, making it more difficult to detect low-abundance proteins. Gives additional information about the protein: A Western blot can confirm the correct protein is being measured by using a number of factors such as protein size.
