In molecular biology and genetics, a blot is a method of transferring large biomolecules (proteins, DNA or RNA) onto a carrier, such as a membrane composed of nitrocellulose, polyvinylidene fluoride or nylon.
The main techniques for visualizing a western blot are colorimetric, chemiluminescence, and fluorescence. Colorimetric and chemiluminescence act by an enzymatic reaction either by horseradish peroxidase or alkaline phosphatase (also used in ELISA).
A western blot is a laboratory method used to detect specific protein molecules from among a mixture of proteins. This mixture can include all of the proteins associated with a particular tissue or cell type.
A Western blot test is typically used to confirm a positive HIV diagnosis. During the test, a small sample of blood is taken and it is used to detect HIV antibodies, not the HIV virus itself.
The currently licensed Du Pont Western blot test specifies that the test result should be interpreted as positive only when the detected bands include p24 and p31, and gp41 or gp120/160 (12) (see Table 2).
Western blot analysis is well suited for evaluating levels of protein expression in cells, seeing how a particular protein responds to a given treatment, and identifying protein-protein interactions.
In reading this western blot, note that each condition (i.e., cell line) is loaded in a column. The relative level of a given protein is evaluated across its respective row. For example, this gel shows that Cathepsin D is much more prominent in the Capan-2 cell line than the AsPC-1, BxPC-3, or Capan-1 cell lines.
The ladder establishes standard molecular weight bands that are then used to read the relative weight of proteins.[20] Electrophoretic Transfer (Blotting) Blotting is the electrophoretic transfer of gel contents onto a suitable membrane; in a western blot, the contents are proteins.
