hello this is Sean Taylor field application scientist manager for bio-rad in Canada and Iamp;#39;d like to demonstrate now how to perform density metric analysis of gel or blot images using image lab software so what we have before us here is a file that contains a 1 amp;amp; 2 dilution series of a protein lysate diluted down from about 60 micrograms loading in 1 amp;amp; 2 so 30 15 7.5 all the way down to 6 to the sixth Lane and weamp;#39;re going to try and quantify using image lab the density of these more prominent bends that we see here and maybe a less prominent band here so to do density metric analysis with the software you use these toolbars so you have a tool box on the side here and Iamp;#39;m only going to demonstrate a couple of the tools in this module just to focus in on density metric analysis so it starts with the image tools and the image tools are used strictly to straighten an image or to rotate or flip the image vertically horizontally the custom rotat

In order to make a permanent record, or to get an objective measure of the signal generated on a Western blot, a densitometer is used to scan the blot or film. Imaging software is then used to compare the signal generated by the bands detected on the Western blot.
To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
Western Blot Presentations Label every lane (optional: use numbers and have a number key below) Label ladder band sizes. Label band(s) of interest with protein name. Label control band with protein name.
TIFF 16 bit is 16 bits per channel. JPEG is 8 bits per channel. So: 16 x 3 = 48 bit.
Western blotting is a laboratory technique used to detect a specific protein in a blood or tissue sample. The method involves using gel electrophoresis to separate the samples proteins. The separated proteins are transferred out of the gel to the surface of a membrane.
Western blots and gels should be submitted as separate files and not embedded in the text. For initial submission, several file types are acceptable, but TIFF or PDF files are preferred. For publication, line art should be 1000 dpi. Color images should be 300 dpi at the final print size.
Open the blot image and the ladder image and convert both images to 8 bit (Image Type 8-bit) Generally invert the blot image so that bands are black and background is white (Edit Invert) Adjust both the blot image and the ladder image (Image Adjust Brightness/Contrast) so that they look good to you.
