- [Narrator] Western blotting, also called immunoblotting, allows researchers to determine levels of protein expression in a cell or tissue extract through antibody binding to a specific protein of interest. Although this technique is widely used and accepted, problems can occur that lead to suboptimal results, such as low signal or high background, resulting in blots that are difficult to interpret or quantitate. This is the second video in our two-part series on western blotting. The first video, Western Blotting Protocol, provides a comprehensive western blot procedure used in-house by cell-signaling technology scientists to validate our antibodies. This second video provides a guide to help you troubleshoot, suggesting tips to help diagnose problems, and providing solutions to ensure you get the expected results in the shortest amount of time. - Before we begin, itamp;#39;s important to note that the single biggest contributing factor to western blot success is the quality of the

In this technique a mixture of proteins is separated based on molecular weight, and thus by type, through gel electrophoresis. These results are then transferred to a membrane producing a band for each protein. Western blot (immunoblot) testing identifies IgM and IgG antibodies against specific B burgdorferi antigens. It is considered positive if it detects at least 2 of a possible 3 specific IgM bands in the first 4 weeks of disease or at least 5 of 10 specific IgG bands after 4 weeks of disease (Table 4 and Figure 2).
For a positive control a known source of target protein, such as purified protein or a control lysate is used. This helps to confirm the identity of the protein, and the activity of the antibody. A negative control is a null cell line, such as -actin, is used as well to confirm that the staining is not nonspecific.
Western blotting is frequently used for the confirmatory medical diagnosis of infectious diseases such as Lyme disease, HIV infection, bovine spongiform encephalopathy (BSE), hepatitis C infection, syphilis, inflammatory muscle conditions such as myositis, and certain autoimmune disorders (e.g., paraneoplastic disease)
Interpretation of the results of a dot blot or a slot blot hybridization is relatively straightforward. If hybridization has occurred, a signal is generated in the specific spot. Therefore, a simple yes or no interpretation is usually given. No information is available about the size of the hybridizing fragments.
