To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
Bands from IgG heavy (~50 kD) and light (~25 kD) chains often mask or obstruct the bands of interest in a western blot of immunoprecipitated proteins.
To obtain linear signals with the majority of western blots, we recommend loading smaller amounts of protein sample between 1 and 10 g per well. To avoid under- or overloading samples, determine the protein concentration of each sample prior to electrophoresis with a compatible protein assay.
To perform Western blot normalization using a single protein as a control, the blot is probed with a primary antibody specific for the protein of interest, and one directed against a normalization control. Ideally, the normalization control is a protein that is present at constant levels in every sample.
For best results, the optimal amounts of antibody should be empirically determined. But a general rule is to add 2 to 10 micrograms of antibody per 500 micrograms of lysate. If you are using neat antisera, or an IgG fraction (such as protein-A purified antibody), greater amounts of antibody are likely to be required.
To correctly dilute secondary antibodies for western blotting it is recommended to first make an antibody working stock solution of 1 g/mL and dilute further for the final antibody incubation solution. Recommended dilutions for use with Thermo Scientific ECL substrates and Alexa Fluor Plus conjugates are below.
A typical starting point for western blotting is 1/1000, and this was very frequently an adequate dilution. When not 1/1000, most often the recommended dilution was lower, at 1/250 or 1/500.
Avivas Western Blot Conditions: Typical antibody concentration range = 0.2 - 5.0 ug/mL, or begin with a 1:1000 dilution of antibody reconstituted at 1 mg/mL.
