Dot blot is a technique for detecting, analyzing, and identifying proteins. This technique is similar to western blot, but protein samples are not separated using electrophoresis; instead, proteins are spotted through circular templates directly onto the membrane or paper substrate.
Following are some of the docHub drawbacks of this method. The dot blot cannot determine the molecular weight of the recombinant protein as the samples are not resolved. It can only show the proteins presence in the cell lysate.
Dot blotting is an ideal technique for quickly assessing the levels of a target antigen across many samples at once. Also, it is a popular method for epitope mapping and screening antibodies for target specificity.
Ensure no air bubbles are trapped by rolling them off using a glass rod. Place a stack of paper towels on this, followed by weight (such as glass slab). Let this assembly stand overnight for complete transfer of the DNA fragments. Transfer of DNA fragments up to 15kb takes about 18 hours.
The method can be used on both dry and wet Immobilon PVDF membranes, enabling the analysis in about 2 hours, as opposed to over 4 hours for the standard method. Important: When using a dry membrane, the blot must be thoroughly dry before beginning rapid immunodetection (refer to Membrane Drying Methods).
Limitations of dot-blotting: The dot blot cannot determine the molecular weight of the recombinant protein as the samples are not resolved.
Dot Blot Assay Protocol Blot (10 l) of different concentrations of cell lysates onto the membrane. Blot 10 l of 100 g/ml of primary antibody onto membrane. Incubate the membrane for 1 hour at room temperature.
