MBP is known to docHubly enhance the solubility of many proteins, resulting in higher yield. Whereas, His-tagging is widely employed for the purification of recombinant target proteins via immobilized metal affinity chromatography (IMAC).
Compared to GST, MBP is larger so it may be a less suitable tag for a larger protein of interest (see more below). However, MBP does not dimerize, and the eluting molecule is not a reducing agent. On the other hand, GST is more widely used in downstream applications compared to MBP.
Maltose binding protein (MBP) is a common protein expression tag, as it is known to docHubly enhance the solubility of many proteins. MBP is one of the most well-known and accomplished means of tagging proteins expressed in microbes.
Recombinant E. Coli MBP produced in E. Coli is a single, non-glycosylated polypeptide chain containing 410 amino acids (27-392 a.a) and having a molecular mass of 44.9kDa. MBP is fused to a 24 amino acid His-tag at N-terminus purified by proprietary chromatographic techniques.
Protein tags are peptide sequences genetically grafted onto a recombinant protein. Tags are attached to proteins for various purposes. They can be added to either end of the target protein, so they are either C-terminus or N-terminus specific or are both C-terminus and N-terminus specific.
MBP is a natural affinity tag that binds to amylose resin and it can be utilized for single-step affinity purification by binding to cross-linked amylose. MBP tag-fused recombinant proteins bound to immobilized amylose are typically eluted under non-denaturing conditions using maltose [usually 10 mM].
Advantages of MBP-tagged Proteins: MBP-tagged proteins are usually able to achieve high yields in the cytoplasm of E. coli since the juxtaposition leads to efficient translation initiation. 2. MBP tag can be used to keep its passenger proteins from proteolytic degradation.
TEV Protease It is the most frequently used protease to remove tags such as GST-tag, MBP-tag, and the Streptavidin tag. The molecular weight (MW) of TEV protease is around 27 KDa.
