In molecular biology and genetics, a blot is a method of transferring large biomolecules (proteins, DNA or RNA) onto a carrier, such as a membrane composed of nitrocellulose, polyvinylidene fluoride or nylon.
Reduce and denature the samples by boiling the lysates in Laemmli Buffer at 95-100˚C for 5 minutes. Note: This step should only be skipped if the antibody datasheet recommends non-reducing or non-denaturing conditions.
Before SDS-PAGE, protein samples should undergo a thorough denaturation to lose all advanced confirmations and only present a linear primary structure. To do so, ionic detergent SDS, elevated temperature, and reducing agent are applied. The protein structure is sensitive to both SDS and elevated temperature.
For a routine Western blot, it is recommended to run the gel in reducing/denaturing conditions. For this, the lysate must be boiled in sample buffer at +95-100C (5 minutes) or at +70C (10 minutes). Then, samples can be immediately loaded on a gel or stored at -20C for later analysis.
Thus, it is necessary to unfold or denature the protein to enable access to the antibody. Denaturing is performed by briefly boiling the sample in a loading buffer containing SDS. The most common loading buffer for SDS-PAGE gel electrophoresis is 2X Laemmli buffer.
Heating the sample at 100C in SDS-containing buffer can result in proteolysis. We recommend heating samples for denaturing electrophoresis (reduced or nonreduced) at 70C for 210 minutes for optimal results. Do not heat the samples for non-denaturing (native) electrophoresis.
Blotting is the process of transferring macromolecules (proteins, nucleic acids, etc.) from a gel to the solid surface of an immobilised membrane to detect the molecules that have been transferred. Identification of specific sequences of DNA, RNA, and proteins is important for various studies in Molecular biology.
Protein samples prepared for SDS-PAGE analysis are denatured by heating in the presence of a sample buffer containing 1% SDS with or without a reducing agent such as 20mM DTT, 2-mercaptoethanol (BME) or Tris(2-carboxyethyl)phosphine (TCEP).
