To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
to cause something to disappear, or to remove something unpleasant from your thoughts: A dark cloud blotted out the sun. He blots out the painful memories by keeping very busy.
Western Blot Presentations Label every lane (optional: use numbers and have a number key below) Label ladder band sizes. Label band(s) of interest with protein name. Label control band with protein name.
Western blotting begins with separating a mixture of proteins on a gel, transferring them to a membrane, and detecting one or more targets of interest with labeled antibodies. An image of the membrane is then capturedeither on to film or using a digital imagerto allow for analysis.
Figures should be provided as individual PDF files with one file per figure. Other acceptable file formats for Western blots and gels are TIFF, EPS, AI, and PSD. Images should be captured at a minimum of 300 dpi. A figure with both line art or text and photographs should be 600 dpi.
Gently mark molecular weight ladder bands with a pencil for size detection. If all blue molecular weight markers were used, this step can be omitted as the bands of all blue markers will be visible after detection when used in conjugation with the Blue Marker Antibody.
Chemiluminescence detection is the most common approach for visualizing protein bands on a Western blot and requires exposing the membrane to X-ray film or a digital charge-coupled device (CCD) imager. Both blot visualization methods are commonly used.
