hey guys welcome back I figured Iamp;#39;d do a quick video on how I guide coat and block sand uh primer panel and getting it ready for paint um I always start with 180 grit and work my way up to 600. I like to start with 180 grit because it just cuts the primer down fast and flat I like to put a guide coat on first I use a powdered guide coat itamp;#39;s just a guide coat itamp;#39;s basically black chalk what I do is I Shake It Up and I really coat the whole entire panel make sure you get completely coated you can order this on Amazon you can go to your paint store get it they do sell it in a spray can um but just make sure you get it all black colored foreign now this is a pretty easy panel to block sand um the trunk lid on this car is a lot harder because of the shape of it I usually white and Trump lives and hoods they usually like to go you know multiple different directions but on the trunk lid on this car itamp;#39;s kind of hard to do that okay so completely coated with gu

Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk diluted in TBST to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available.
To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
Most laboratories use a range of 2040g of total protein for loading homogenates for western blotting.
The protein component of the blocking solution should be compatible with both the antigen that is being detected and the species of the secondary antibody. Bovine serum albumin (BSA) and non-fat-dried milk are commonly used in blocking solutions as they are both inexpensive and easily available.
Place the blot in PBS and wash for 5 minutes. Place the blot in the secondary antibody solution and incubate with agitation for 30 minutes. Place the blot in PBS (PBS Tablets 524650-1EA) and wash for 5 minutes. Repeat twice with fresh buffer.
Dont over-block. Long blocking incubations, particularly with nonfat dry milk at 2% or higher, can cause loss of target protein from the membrane (J.
Check protein transfer by staining the blot with Ponceau S for 1 min, then completely destain the blot by washing with distilled water. Place blot into blocking solution for 2 hr at RT, or overnight at 4C.
Note on blocking: Soak the blotted membrane in freshly prepared blocking reagent, PBS/3% nonfat dry milk (15gms nonfat milk in 500mLs PBS (PBS Tablets 524650-1EA) for 30 minutes to 2 hours at room temperature with constant agitation. Membranes may also be blocked with PBS/3% nonfat dry milk overnight at 4C.
