Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk diluted in TBST to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available.
The principles of western blotting are equal loading of proteins, separation of proteins by molecular weight, electrophoretic transfer to a suitable membrane, and probing of antibodies. Proper sample preparation for subsequent electrophoresis is crucial for downstream analysis.
The term western blot was given by W. Neal Burnette in 1981, although the method itself was independently invented in 1979 by Jaime Renart, Jakob Reiser, and George Stark at Stanford University, and by Harry Towbin, Theophil Staehelin, and Julian Gordon at the Friedrich Miescher Institute in Basel, Switzerland.
Skimmed milk powder is particularly good at blocking the membranes vacant surface area, as its wild mix of very varied proteins provides particularly effective cover. With strong binding antibodies, milk powder works really well, says Ruth.
Western blotting is a laboratory technique used to detect a specific protein in a blood or tissue sample. The method involves using gel electrophoresis to separate the samples proteins. The separated proteins are transferred out of the gel to the surface of a membrane.
Non-fat dry milk (NFDM) blocking buffer is ideal for saturating excess protein-binding sites on membranes for Western blotting. The primary proteins in milk are casein and whey. Typically, 1-5% NFDM is sufficient for most Western blots.
But for antibodies with low binding strength, or their target proteins may be scarcely present in the cell, it is always better and safer to use BSA only because BSA is a single purified protein. In contrast, NFM constitutes a few mixed proteins that may hinder the actual antibody binding.
To prevent non-specific binding of detection antibodies during the steps following transfer, unoccupied sites on the membrane surface must be blocked. Blocking buffer formulations vary widely and may contain milk, normal serum, or highly purified proteins to block free membrane sites.
