The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue. It can stain cells blue or blue-green (basophilia, cyanophilia), pink (acidophilia, eosinophilia), orange (orangeophilia), or indeterminate (gray-blue).

Pap being a differential staining technique imparts red to pink color to the keratinized mature cells and blue to green color to the immature or intermediate cells that are metabolically active. In pap smears, Light Green SF confers a blue staining for the cytoplasm of active cells such as columnar cells, parabasal squamous cells, and intermediate squamous cells. It usually comes as a disodium salt. Its maximum absorption is at 630 (422) nm.
Light green SF is generally used as a cytoplasmic counterstain with nuclear stains like safranine and Feulgen stain. In histopatology, it is used for collagen fiber staining in Massons trichrome, Papanicolaou cytological polychrome stains, and Twort stain for microorganisms in tissue sections. The stain should result in cells that are fairly transparent so even thicker specimens with overlapping cells can be interpreted.
