To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot. Blotting is the process of transferring macromolecules (proteins, nucleic acids, etc.) from a gel to the solid surface of an immobilised membrane to detect the molecules that have been transferred. Identification of specific sequences of DNA, RNA, and proteins is important for various studies in Molecular biology.
It detects viral antigens (proteins usually on the surface of viruses) using antibodies against those proteins. A positive Western blot indicates the presence of viral antigen which very often means live virus in our patient. That patient may have an ongoing viral infection.
The Cell Cycle western blot cocktail (ab136810) is designed to provide a rapid assessment of cell cycle distribution based on molecular markers for the G1/S and M phases of the cell cycle.
A speckled or spotty background can be caused by foreign material adhering to the membrane, antibody aggregation, or dirty scanner or cassette surfaces. All are preventable by fastidious handling procedures.
Gently mark molecular weight ladder bands with a pencil for size detection. If all blue molecular weight markers were used, this step can be omitted as the bands of all blue markers will be visible after detection when used in conjugation with the Blue Marker Antibody. A DNA marker (also known as a size standard or a DNA ladder) is loaded into the first well of the gel. The fragments in the marker are of a known length so can be used to help approximate the size of the fragments in the samples. The prepared DNA samples are then pipetted into the remaining wells of the gel.
Protein molecular weight markers, sometimes referred to as protein standards or protein ladders, are used to estimate the molecular weight of proteins of interest and to monitor the progress of electrophoretic separation or transfer in Western blotting.
These markers are intended for use as a size standard when performing SDS-PAGE (sodium dodecyl sulfate-polyacrylamide gel electrophoresis) for estimation of the molecular weight of the protein of interest. Note that they are not stained.
