Welcome to the Novus Visual Protocol Series. In this video we will learn how to perform all phases of a Western Blot using the most common methods for this assay. Before we can start preparing the blot we must first prepare our sample lysate. In this example we will prepare a protein lysate from cultured cells. Here we wash the cells twice with ice cold PBS and enough lysis buffer to cover the cells. The choice of lysis buffer depends largely upon the localization of your protein of interest. We scrape the cells and transfer the cell solution on a centrifuge tube placed on ice. In order to solubilize membrane bound proteins, we will require stronger extraction detergents compared to isolated cytoplasmic proteins. In this example we are using a standard RIPA buffer, which is a common buffer for obtaining maximum protein yield. While extracting proteins from all cellular localizations, it is very important to include protease inhibitors in your lysis buffer which will prevent degradation

In a traditional western blot (indirect labeling), protein samples are first resolved by SDS PAGE and then electrophoretically transferred to the membrane. After a blocking step, the membrane is probed with a primary antibody that was raised against the antigen in question.
SDS-PAGE is typically used for western blotting, where proteins are denatured and reduced to obtain their primary structure. Coating with SDS molecules (Sodium dodecyl sulfate) imparts a relative negative charge proportional to their molecular weight, allowing separation of the protein by size only.
SigmaMarker protein markers are designed for use with SDS-PAGE and cover the range of molecular weights common to most proteins or their subunits. These high and low wide range markers are lyophilized with sample buffer so that they are ready-to-use after reconstitution with deionized water.
Show or hide formatting marks Show invisibles: Choose View Show Invisibles (from the View menu at the top of your screen). Note: The View button in the toolbar doesnt have this command. Hide invisibles: Choose View Hide Invisibles.
Show or Hide Formatting Symbols Click Home tab. Click Show/Hide button. The hidden formatting symbols are now shown. These symbols represent spaces, new paragraphs, page breaks, and other bits of text and page formatting. Click Show/Hide button again.
Quickly toggle all formatting marks (When you point your mouse at the button, the tooltip says Show/Hide ). Keyboard shortcut CTRL+SHIFT+*.
Toggle visibility of non-printing symbols by clicking the button on the Home ribbon (or pressing Ctrl-Shift-8 (Windows) or Command-8 (Mac)).
