Long kept confidential for psychologists and their patients, the mysterious images were said to draw out the workings of a persons mind. But what can inkblots really tell us, and how does this test work? Invented in the early 20th century by Swiss psychiatrist Hermann Rorschach, the Rorschach Test is actually less about the specific things we see, and more about our general approach to perception. As an amateur artist Hermann was fascinated by how visual perception varies from person to person. He carried this interest to medical school, where he learned all our senses are deeply connected. A speckled or spotty background can be caused by foreign material adhering to the membrane, antibody aggregation, or dirty scanner or cassette surfaces. All are preventable by fastidious handling procedures.
Western Blot Marker Pen ab166858 is used to annotate the blot and/or to mark the positions of (pre-stained) molecular weight standard markers on a PVDF or nitrocellulose membrane during western blotting. The pen is used after the transfer of proteins from the gel to the membrane.
Problem: Blotchy or patchy background A blotchy background can be due to a dry membrane or insufficient washing; both problems are readily solved by ensuring that the blot is immersed at all times.
Observation: Patchy uneven spots all over the blot Make sure membrane is fully immersed during washes and antibody incubations. Gently remove any air bubbles. Especially during transfer. Ensure uniform agitation by placing on a rocker/shaker.
Different blots are used to identify the existence of one particular target molecule, such as DNA, RNA or protein, in an intricate mixture of associated molecules. Blotting is the process of transferring macromolecules (proteins, nucleic acids, etc.)
The Western blot test separates the blood proteins and detects the specific proteins (called HIV antibodies) that indicate an HIV infection.
Bubbles are the result of air being trapped between the membrane and the gel during transfer. Usually bubbling is minor, and if it is just a couple of bubbles like in the blot above, then I simply use a gel pen to circle them on the blot.
Blotchy, Flecked, Or Dirty Background These artifacts are most commonly the result of uneven coating of buffer or antibody, the membrane drying out, or aggregates forming in the antibody or blocking buffer.
