Western blotting is an important technique used in cell and molecular biology. By using a western blot, researchers are able to identify specific proteins from a complex mixture of proteins extracted from cells.
Traditionally, protein signal on blots was generated colorimetrically or using chemiluminescent substrates and captured using film. Only rough estimates of protein quantity could be inferred by eye, but converting that film image into a digital image allowed more accurate analysis of the data.
Blotchy, Flecked, Or Dirty Background These artifacts are most commonly the result of uneven coating of buffer or antibody, the membrane drying out, or aggregates forming in the antibody or blocking buffer.
Chemiluminescent western blot signal can be captured with X-ray film, charge-coupled device (CCD) camerabased digital imaging instruments, and phosphorimagers that detect chemiluminescence.
Western blotting begins with separating a mixture of proteins on a gel, transferring them to a membrane, and detecting one or more targets of interest with labeled antibodies. An image of the membrane is then capturedeither on to film or using a digital imagerto allow for analysis.
SDS binds strongly to proteins, with approximately one detergent molecule binding to two amino acids when SDS is present at 0.1% (1,2). When boiled with SDS, proteins gain a negative charge in proportion to their molecular size, and thus travel in the acrylamide gel ing to their molecular sizes. What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
SDS-PAGE is typically used for western blotting, where proteins are denatured and reduced to obtain their primary structure. Coating with SDS molecules (Sodium dodecyl sulfate) imparts a relative negative charge proportional to their molecular weight, allowing separation of the protein by size only.
