The Southern blot is used to verify the presence or absence of a specific nucleotide sequence in the DNA from different sources, and to identify the size of the restriction fragment that contains the sequence. In this procedure, the DNA is isolated from each source and then digested with a specific restriction enzyme. The DNA restriction fragments are then loaded onto an agarose gel, and the fragments separated by electrophoresis ing to size, with the smaller fragments migrating faster than larger fragments. The fragments are made visible with ethidium bromide. The DNA is then transferred from the fragile gel to a nylon filter. Next, the radioactively labeled nucleic acid probe is added. The probe binds to complementary DNA segments. Note that the DNA segment being probed is not present in organism B. To detect the position of the radioactive probe, the nylon membrane is covered with an X-ray film. After development, the positions of the probe become visible.

What is line blot? Line blots are immunoassays in which antigens are placed precisely on nitrocellulose as narrow lines. The line blot is an immunoassay for monoclonal and other antibodies. It is an application to the serotyping of gram-negative bacteria.
What is ANA blot test? This test is done in cases where an autoimmune disease is suspected; an ANA By IF is first done. The standard algorithm is to do an ANA by IF, and if this is positive, to follow up with a monospecific Line BLOT. i.e This Test is used to confirm an antigen /Pattern detected by ANA by IIFT (ANAIF).
ANA test results are most often reported in 2 parts: the level or titer and the pattern. Titres are reported in ratios, most often 1:40, 1:80, 1:160, 1:320, and 1:640. Some, but not all labs will report a titre above 1:160 as positive. Patterns that are reported include, homogeneous, speckled, centromere, and others.
In contrast, antinuclear antibodies often attack your bodys own tissues specifically targeting each cells nucleus. In most cases, a positive ANA test indicates that your immune system has launched a misdirected attack on your own tissue in other words, an autoimmune reaction.
Your provider will use your ANA test results along with other tests and information about your health to make a diagnosis. A negative result on an ANA test means that antinuclear antibodies were not found in your blood, and youre less likely to have an autoimmune disorder.
A positive ANA reading may indicate a need for follow up blood tests, physical exam, and a review of history and symptoms. Additionally, some medications, viruses, and conditions (such as cancer) can cause a positive ANA.
