In a traditional western blot (indirect labeling), protein samples are first resolved by SDS PAGE and then electrophoretically transferred to the membrane. After a blocking step, the membrane is probed with a primary antibody that was raised against the antigen in question.
A typical blotting experiment involves these 4 steps: gel electrophoresis of the sample. transferring or blotting the electrophoresed samples from the gel to a membrane. treating the membrane with a reagent(s) (probe) that binds to the target molecule and detects it on the membrane.
To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
All western blot samples have three elements: protein extract, cell lysis buffer, and Laemmli (sample) buffer. Protein extract is normalized with cell-lysis buffer to the desired protein concentration, and an equal volume of Laemmli (sample) buffer is added.
Western Blot Presentations Label every lane (optional: use numbers and have a number key below) Label ladder band sizes. Label band(s) of interest with protein name. Label control band with protein name.
Ideally, control samples should be run on the same blot or gel as the experiment. Relevant controls must be included in each figure. A figure containing multiple blots or gels should clearly display the different images as separate panels and this should also be noted in the figure legend.
Figures should be provided as individual PDF files with one file per figure. Other acceptable file formats for Western blots and gels are TIFF, EPS, AI, and PSD. Images should be captured at a minimum of 300 dpi. A figure with both line art or text and photographs should be 600 dpi.
What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
