Antibody labeling, or the attachment of a specific tag to an antibody to aid in detection or isolation/purification of a protein, is an important technique. Labeled antibodies are essential for immuno-based assays such as Western blots, ELISAs, flow cytometry, immunohistochemistry (IHC) and immunofluorescence (IF).
Site-specific antibody labeling ensures that the label does not interfere with antigen binding. This becomes particularly important when attaching large biomolecules to the antibody (e.g. enzymes), which can sterically prevent antigen binding.
Covalent conjugation via NHS ester groups is the most common antibody labeling method, which involves attaching the label of interest to the amine groups of lysine residues. The NHS ester method uses fluorescent dyes with a reactive N-Hydroxysuccinimide (NHS) group (Fig. 3).
Direct labelling (left) uses an antibody tagged with a fluorescent marker to label target antigens while indirect labelling (right) uses secondary antibodies tagged with the markers to target the primary antibody that has bound to the target antigen.
Labels used to conjugate antibodies include fluorescent dyes, small molecules (eg, biotin), enzymes (horseradish peroxidase (HRP) or alkaline phosphatase (AP)), proteins (eg, R-PE, APC, streptavidin), latex or gold nanoparticles, and oligonucleotides.
Western blotting technique is used to identify the isolated protein. It is also known as immunoblotting, because an antibody is used to specifically detect its antigen. ​Harry Towbin in 1979 developed this technique while experimenting in a laboratory in Switzerland.
A label can be an enzyme, a fluorophore or colloidal gold. The enzyme can produce a chromogenic reaction making identification possible. The fluorescent microscope is used to view a fluorophore label. The electron microscope is used to view colloidal gold.
