We can do this by taking the material from the sample and running it on a gel, and then transferring the resolved proteins onto a special piece of membrane--of paper, if you will--and then probe that paper with an antibody to the specific protein of interest.
There are six steps involved in western blot, including sample preparation, gel electrophoresis, proteins transfer, blocking, antibody incubation, and proteins detection and visualization.
Using ImageJ to Quantify Bands Save gel image and adjust to be vertically oriented. Using a rectangular box (box tool)select entire lane. Select lane either by selecting Analyze-Gels-Select First Lane or press CTRL-1.
To know how to analyze western blot data, Look for the sizes of the bands. These will be represented by a number, either followed by kDa or preceded by p. This is the size of the protein which has been detected and is the scale on which the proteins are separated in a Western blot.
A western blot image is made up of pixels, which contain information about how much signal was collected at each location in the image. A digital image of a blot can be thought of as data in three dimensions.
By using a western blot, researchers are able to identify specific proteins from a complex mixture of proteins extracted from cells.
The first step in a western blot is to prepare the protein sample by mixing it with a detergent called sodium dodecyl sulfate, which makes the proteins unfold into linear chains and coats then with a negative charge.
Select AnalyzeGelsSelect First Lane (or press 1) and Lane 1 selected will be displayed in the status bar. ImageJ will also duplicate the image and outline and label the lane if Outline Lanes is checked in the AnalyzeGelsGel Analyzer Options.
