hi im alicia and today im going to show you how to quantify bands for your western blots so first open the file up and itll look something like this as a just scanned image i like to edit the contrast so that you can see all the faint bands too you can manually add all the lanes which is what i like to do or you could do automatic and then fiddle around with this to make it fit nicely i sped this up a bit because it takes a while next you have to make sure each lane covers the whole band at the particular molecular weight this takes a while too so dont worry just take your time with it alright next then you add bands for each lane and it should be at about the same molecular weight go to lane profile to adjust for noise background noise and then you want to make sure that the green peak the green is the volume of the whole mountain type thing so drag it out a little bit if it goes a little further you want to make sure that youre counting for at the very end of the peak sometimes
