IBI TWEEN 20 can be used as washing agent immunoassays such as Western Blots and ELISAs. It helps prevent non-specific antibody binding. In these applications it is typically dissolved in Tris-Buffered saline (TBS) or Phosphate Buffered saline (PBS) at dilutions of 0.05% in 1X or 0.5% in 10X solution.

Dot blot is a technique for detecting, analyzing, and identifying proteins. This technique is similar to western blot, but protein samples are not separated using electrophoresis; instead, proteins are spotted through circular templates directly onto the membrane or paper substrate.

TBS Tween20 is one of the most commonly used biological buffers for plate based immunoassays and Western blotting. It is isotonic, non-toxic to cells and contains a non-ionic detergent.

TBST buffer is used in Western Blot to wash away non-specific binding on the membrane, this buffer contains 0.1% Tween20. Tween20 is a non-ionic detergent that increases the buffers ability to elute and reduce non-specific binding of antibodies.

Why do we use TBST in western blot?

For example, PBS interferes with alkaline phosphatase (AP)-conjugated secondary antibody and in this case, you should use TBS instead. TBS blocking buffers are also the best choice for detecting phosphorylated protein molecules with phospo-specific antibodies.

What does TBST do in a western blot?

Blocking is a very important step of western blotting, as it prevents antibodies from binding to the membrane nonspecifically. Blocking is often made with 5% BSA or nonfat dried milk diluted in TBST to reduce the background. Nonfat dried milk is often preferred as it is inexpensive and widely available.
