Western blot is a technique that is very useful for protein detection as it allows the user to quantify the protein expression as well.
The western blot (also known as immunoblotting, because an antibody is used to identify its antigen) is a widely used analytical method often used by researchers to detect the presence of a protein in a given sample (Towbin, Staehelin, Gordon, 1979) (Fig.
Here, we review four popular methods: Northern blot analysis, nuclease protection assays (NPA), in situ hybridization, and reverse transcription-polymerase chain reaction (RT-PCR). In theory, each of these techniques can be used to detect specific RNAs and to precisely determine their expression level.
techniques are used to detect and analyze three types of biological macromolecules: DNA, RNA and proteins. Results of a blotting experiment tell you whether a macromolecule of a specific sequence is present in your sample or not.
RNA purity is determined by measuring the 260/280 and 260/230 ratios using a spectrophotometer. Excessive absorbance at 280 indicates the presence of protein in your sample while excessive absorbance at 230 may indicate the presence of residual phenol in your sample.
A northern blot is a laboratory method used to detect specific RNA molecules among a mixture of RNA. Northern blotting can be used to analyze a sample of RNA from a particular tissue or cell type in order to measure the RNA expression of particular genes.
This technique is called electrophoresis. The investigator then transfers the separated RNAs to a nitrocellulose paper or nylon membrane filter, and incubates the RNA-containing filter in a solution containing a radioactive single-stranded DNA fragment from a particular gene (Figure 4.14A).
Northern blot is a laboratory analysis method used to study RNA. Specifically, purified RNA fragments from a biological sample (such as blood or tissue) are separated by using an electric current to move them through a sieve-like gel or matrix, which allows smaller fragments to move faster than larger fragments.
