In this video, I will show you how to perform a Western blot. The purpose of a Western blot is to specifically look at a single protein and visualize it on a camera. The steps included in running a Western blot are a protein gel, which can be found in a different video. But using the product of this protein gel, the proteins themselves are transferred onto a membrane. A primary antibody is added to the membrane to specifically bind to your protein of interest. A secondary antibody binds the first antibody, and the secondary antibody usually contains some sort of fluorophore, which we are then able to see when placing everything into a camera system. To run a Western blot, the first step after running the protein gel is to assemble the Western sandwich. The Western sandwich consists of three sheets of Whatman paper, the gel, a nitrocellulo

The lanes on my Western blot are smeared Smeared lanes are almost always due to poor sample preparation, which can lead to degradation, protein aggregation, and over-loading.
Blotchy, Flecked, Or Dirty Background These artifacts are most commonly the result of uneven coating of buffer or antibody, the membrane drying out, or aggregates forming in the antibody or blocking buffer.
Different blots are used to identify the existence of one particular target molecule, such as DNA, RNA or protein, in an intricate mixture of associated molecules. Blotting is the process of transferring macromolecules (proteins, nucleic acids, etc.)
A blotchy background can be due to a dry membrane or insufficient washing; both problems are readily solved by ensuring that the blot is immersed at all times. Additional handling precautions should be taken to prevent accidental contamination from common laboratory sources.
Sometime, not washing the blot enough can give this kind of problems. I would advice you to wash your blot for at least a total 30 mins (3 changes with 10mins each) after keeping in the primary. washing the blot before addition of secondary antibody may lead to unspecific binding.
What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
Blocking is critical for clean blots. Blocking agents such as non-fat dry milk or BSA occupy non-specific binding sites resulting in lower background. Check to see if there is a preferred blocking agent and concentration for your antibody. A 1-5% blocking solution is usually recommended.
In molecular biology and genetics, a blot is a method of transferring large biomolecules (proteins, DNA or RNA) onto a carrier, such as a membrane composed of nitrocellulose, polyvinylidene fluoride or nylon.
