IP-Western analysis remains a popular technique for identifying protein-protein interactions and identifying unknown proteins in a multi-protein complex. The steps include cell lysis, formation of the antibody-antigen (immune) complex, precipitation of the immune complexes, and analysis by Western blotting.
Major types of blotting techniques include Southern blotting (for DNA), Northern blotting (for RNA), Western blotting (for proteins), Southwestern blotting (for DNA-protein interactions), and Eastern blotting (for post-translational modifications of proteins).
However, the main difference that has to be noted is the molecule that each technique is meant to identify. Southern blot detects specific DNA sequences, Northern blot detects particular RNA sequences, and Western blot detects specific proteins. All these techniques are complementary.
How long do you incubate primary antibody in a Western Blot? It depends on the binding affinity of the antibody to the target protein, usually 2 hours at room temperature incubation or 4C incubation overnight.
