FLAG Tag Protein Purification Anti-FLAG M2 Magnetic Beads are used to purify recombinant fusion-tagged protein using a magnetic rack or platform to separate beads and isolate protein for rapid processing.
A Flag-tagged protein can be immunoprecipitated from a crude cellular extract using an antibody bound to the Flag-tag, an anti-Flag antibody. As Flag is a registered trademark, anti-Flag antibodies are often called DYKDDDDK antibodies.
As mentioned above, 3x-Flag-tag is longer than 1x Flag-tag and has more charged amino acids. Negatively and positively charged amino acids potentially interfere more strongly with the POI and may influence protein expression. For detection in Western blot, 1x Flag-tag should be sufficient.
Like the original FLAG tag, 3xFLAG is hydrophilic, contains an Enterokinase cleavage site, and is relatively small. Therefore, the risk of altering protein function, blocking other epitopes, or decreasing solubility is minimized.
The increased length of the 3x Flag-tag increases the affinity of the anti-Flag antibody/affinity reagents. 3x Flag-tag is often used in tandem purification and protein purification. As mentioned above, 3x-Flag-tag is longer than 1x Flag-tag and has more charged amino acids.
Anti-FLAG Western blot antibodies are immunoblotting reagents for the qualitative or semi-quantitative detection of FLAG-tagged proteins and peptides. FLAG is an octapeptide epitope tag with the amino acid sequence DYKDDDDK that can be added to the N-terminus, C-terminus, or within a recombinant protein.
The 3x Flag-tag synthetic peptide contains 23 amino acid residue. Eight amino acids at the C-terminus make up the classic FLAG sequence Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys. Additionally the motif Asp-Tyr-Lys-Xaa-Xaa-Asp is repeated 3-times in the peptide sequence.
The Flag-tag is a short peptide tag and is commonly used as 1x Flag (amino acid sequence: DYKDDDDK), or 3x Flag (DYKDHD-G-DYKDHD-I-DYKDDDDK). It can be cloned to a protein of interest (POI) either N- or C-terminally.
