Anti-FLAG antibodies are reagents that can be used to detect or stain Flag-tagged recombinant proteins. The FLAG epitope tag, is an octapeptide containing the amino acid sequence DYKDDDDK.
The 3xFLAG system is an improvement upon the original system by fusing 3 tandem FLAG epitopes (22 amino acids). Detection of fusion proteins containing 3xFLAG is enhanced up to 200 times more than any other system. Like the original FLAG tag, 3xFLAG is hydrophilic, contains an Ek cleavage site, and is relatively small.
The 3xFLAG system is an improvement upon the original system by fusing three tandem FLAG epitopes for a total of 22 amino acids (Figure 1).
FLAG-tag, or FLAG octapeptide, or FLAG epitope, is a peptide protein tag that can be added to a protein using recombinant DNA technology, having the sequence DYKDDDDK (where D=aspartic acid, Y=tyrosine, and K=lysine).
Like the original FLAG tag, 3xFLAG is hydrophilic, contains an Enterokinase cleavage site, and is relatively small. Therefore, the risk of altering protein function, blocking other epitopes, or decreasing solubility is minimized.
Anti-FLAG Western blot antibodies are immunoblotting reagents for the qualitative or semi-quantitative detection of FLAG-tagged proteins and peptides. FLAG is an octapeptide epitope tag with the amino acid sequence DYKDDDDK that can be added to the N-terminus, C-terminus, or within a recombinant protein.
3x Flag-tag is often used in tandem purification and protein purification. As mentioned above, 3x-Flag-tag is longer than 1x Flag-tag and has more charged amino acids. Negatively and positively charged amino acids potentially interfere more strongly with the POI and may influence protein expression.
