Blotting is a common diagnostic technique used in molecular biology to identify proteins and nucleic acids. This method immobilises the target molecule on a support, such as a nylon or nitrocellulose membrane.
It detects viral antigens (proteins usually on the surface of viruses) using antibodies against those proteins. A positive Western blot indicates the presence of viral antigen which very often means live virus in our patient. That patient may have an ongoing viral infection.
These filter papers are smooth and suitable for use with alcohol and other organic solvents involved in protein transfer and nucleic acid blotting. They also allow for a uniform flow of buffer through the gel to the transfer membrane in a blotting sandwich.
Blotting papers come pre-cut to completely cover the gels and membranes. Semi-dry transfer setup. In a semi-dry transfer, the stack consists of gel and membrane placed between two pieces of filter paper, all equilibrated in transfer buffer, and this stack is placed directly between two electrode plates.
Nitrocellulose and PDVF (polyvinylidene fluoride) are the most frequently used types of blotting membranes. Nitrocellulose, while less sensitive than PVDF, has lower background noise and is more suitable for detecting proteins of low molecular weight.
Nylon membranes have high mechanical strength, which offers benefits in western blot experiments requiring stripping and reprobing procedures. Nylon membranes consistently provide a higher efficiency of protein transfer SDS gels than nitrocellulose membranes.
PVDF is favored as a material due to its high thermal stability, good chemical resistance, high hydrophobicity, and good mechanical strength. PVDF has a protein binding capacity of 170 to 200 g/cm2, which is higher than that of nitrocellulose and therefore provides greater sensitivity for proteins with low abundance.
Northern blot first uses denaturing gel to separate RNA ing to the size. The RNA is then transferred to a nylon membrane while keeping the same distribution in the gel. After fixing the RNA to the membrane, labeled probe complementary to the gene of interest is then added to hybridize to the immobilized RNA.
