How to blot dot in the Training Evaluation

The Kirkpatrick model of evaluation, developed by Donald Kirkpatrick in the 1950s, is essential for instructional designers and training program evaluators. It is arguably the most popular framework for training evaluation. The model consists of four levels: Level 1 is Reaction, assessing immediate feedback from participants; Level 2 is Learning, measuring the increase in knowledge or skills; Level 3 is Behavior, evaluating the application of learning in the workplace; and Level 4 is Results, which analyzes the overall impact on organizational goals. As one progresses through the levels, the evaluations become more challenging but also more valuable, making this model a fundamental starting point for effective training assessment.

Dot blot also precludes monitoring the expression of a normal and modified target within the same blot. Moreover, because dot blot does not generally include a method for normalization (e.g., detection of a housekeeping protein), accurately comparing protein levels between different samples is not possible. Advantages of Western blotting include the ability to identify specific target proteins, while disadvantages include difficulties in reproducibility and time-consuming procedures. Limitations of dot-blotting: The dot blot cannot determine the molecular weight of the recombinant protein as the samples are not resolved. It can only show the proteins presence in the cell lysate. However, sometimes it is essential to know whether the protein expressed is of the right size (molecular weight).
It involves the application of a small amount of sample directly onto a membrane, which is then probed with a specific antibody or nucleic acid probe to detect the target molecule.
A dot blot (or slot blot) is a technique in molecular biology used to detect proteins. It represents a simplification of the western blot method, with the exception that the proteins to be detected are not first separated by electrophoresis.
As with all techniques, western blotting has its limitations [4,5,11]. The main limitation of western blotting is that it can only be carried out if a primary antibody against the protein of interest is available. The main disadvantage of Western blotting is that this technique requires a specific antibody to a target protein; thus many protein targets cannot be investigated because of the lack of specific antibodies. Dot blots are like Western blots in that they involve antibody-based detection of membrane-bound proteins. However, the main difference between the two techniques is that dot blotting does not require electrophoretic separation.
