Dot and slot blots differ only in the geometry of the blot, a series of spots giving a hybridization pattern that is amenable to analysis by densitometric scanning. Samples are usually applied to the membrane using a manifold attached to a suction device.
A dot blot (or slot blot) is a technique in molecular biology used to detect proteins. It represents a simplification of the western blot method, with the exception that the proteins to be detected are not first separated by electrophoresis.
Dot blot hybridization RNA dot blot is semi-quantitative technique for rapid quantification of relative concentrations of RNA, developed by Kafatos et al. in 1979 [43]. It is mostly used in gene expression assays in different samples.
Interpretation of the results of a dot blot or a slot blot hybridization is relatively straightforward. If hybridization has occurred, a signal is generated in the specific spot. Therefore, a simple yes or no interpretation is usually given. No information is available about the size of the hybridizing fragments.
A major advantage of dot blot over Western blot is that it is much faster. Dot blot is also more cost-effective, since it eliminates the need for materials such as polyacrylamide gels, molecular weight markers, and all of the equipment and buffers required for gel running and protein transfer.
Dot and slot blotting are simple techniques for immobilizing bulk unfractionated DNA on a nitrocellulose or nylon membrane. Hybridization analysis can then be carried out to determine the relative abundance of target sequences in the blotted DNA preparations.
Electrophoretic as well as non-electrophoretic transfer of proteins to membranes was first described in 1979. Protein blotting has evolved greatly since the inception of this protocol, allowing protein transfer to be accomplished in a variety of ways.
