To use the percent area method, open the Dot Plot image in Image J and invert it, making the dots white against a dark background. Employ selection tools to mark background noise and then use the Analyze feature to measure intensity, noting the main intensity value. Deselect the background noise, navigate to Process > Math, and subtract the background using the mean intensity value. Ensure the image has a dark foreground and white background.
