- So in this video, Im gonna be talking about something known as a Southern Blot. So, a Southern Blot basically allows you to visualize a specific piece of DNA that youre interested in. So lets imagine that we have a cup and its filled with DNA. So its got just a whole bunch of DNA inside. And theres just lots and lots of those DNA and lets imagine that Im specifically interested in one gene. So lets imagine that Im interested in Gene A and I want to see if Gene A is inside of this cup. If its inside of this long piece of DNA. Now, in order to figure out whether or not Gene A is inside this cup, basically we have to do this process known as a Southern Blot. And well break it up into a couple of different steps. So Step 1, what were gonna do is were gonna take this DNA and were gonna cleave this. So, take the DNA and cleave it. So, let me draw that out. So, were gonna take this big old strand. General dot blot procedure. The concentration of proteins in crude preparations (such as culture supernatant) can be estimated semiquantitatively using the dot blot method if you have both purified protein and specific antibody against it.
Dot Blot Assay Protocol Use a strip of nitrocellulose membrane. Blot (10 l) of different concentrations of recombinant protein onto membrane. Blot (10 l) of different concentrations of cell lysates onto the membrane. Blot 10 l of 100 g/ml of primary antibody onto membrane.
A dot blot (or slot blot) is a technique in molecular biology used to detect proteins. It represents a simplification of the western blot method, with the exception that the proteins to be detected are not first separated by electrophoresis.
Interpretation of the results of a dot blot or a slot blot hybridization is relatively straightforward. If hybridization has occurred, a signal is generated in the specific spot. Therefore, a simple yes or no interpretation is usually given. No information is available about the size of the hybridizing fragments.
Dot blot hybridization RNA dot blot is semi-quantitative technique for rapid quantification of relative concentrations of RNA, developed by Kafatos et al. in 1979 [43]. It is mostly used in gene expression assays in different samples.
Quantitative dot blot or QDB is a useful technique to quantify the concentration of target proteins in a sample. Once the sample is immobilized on a polymeric membrane, the proteins can be detected via immunoblotting.
There are two built in methods for analyzing a dot blot in ImageJ. The first is to treat each row as a horizontal lane and use ImageJs gel analysis function. The second is to subtract the background and measure the integrated density of each dot.
