This tutorial demonstrates two methods for quantifying dot blood images: the percent area method and the integrated density method. For the percent area method, first, open the dot plot image in ImageJ and invert it to make the dots white against a dark background. Use selection tools to mark background noise, then click "Analyze" and measure the intensity, noting the main intensity of 54.75. Next, deselect the background noise, go to "Process," then "Math," and subtract the background using the mean intensity. To prepare the image for quantification, ensure it has a dark foreground and white background by inverting it again. Finally, use the rectangular selection tool to enclose all dots and proceed to analyze gels for quantification.
